First stop: Run a full system check, you may have a corrupted data file (it happens every now and then due to the constant patching) -- Running Full System Check
Have you recently updated any drivers? Possibly even via Windows Update or Microsoft Update.
If the answer is no then give your Harddrive a defrag, MxO seems to really benefit from a defrag. It may be that ther is problems caused by having certain data fils in too many parts or a file split at the wrong point in the data file when patching occurs.
If that doesn't fix the problem then reinstall MxO from scratch.

In addition to phi's excellent advice it would help to know what the error message is, could you post a screenshot of it?
Could you also post the "Services" , "Startup Programs" and "Windows Errors Reporting" sections from your misinfo.txt file?
